產品概述
胰島素是由胰島β細胞受內源性或外源性物質如葡萄糖、乳糖、核糖、精氨酸、胰高血糖素等的刺激而分泌的一種蛋白質激素。由A、B鏈組成,共含51個氨基酸殘基。能增強細胞對葡萄糖的攝取利用,對蛋白質及脂質代謝有促進合成的作用。胰島素是機體內唯一降低血糖的激素,也是唯一同時促進糖原、脂肪、蛋白質合成的激素。
產品原理
本實驗采用雙抗體夾心ELISA法??勾笫驣nsulin單抗包被于酶標板上,同時加入標本、標準品和生物素化的抗大鼠Insulin抗體,標本、標準品中的Insulin會與加入于孔中的生物素化的抗大鼠Insulin抗體及包被于酶標板上的單抗結合,形成免疫復合物,游離的成分被洗去;加入辣根過氧化物酶標記的親合素,親合素與生物素特異性結合,游離的成分被洗去。加入顯色底物(顯色劑),若反應孔中有Insulin,辣根過氧化物酶會使無色的顯色劑變藍,加終止液變黃。在450nm處測OD值,Insulin濃度與OD450值之間呈正相關,可通過繪制標準曲線求出標本中Insulin濃度。
